Matt Bomer began his television career in 2000 with a role as Ian Kipling on the soap opera All My Children. He appeared on Guiding Light from 2001 to 2003 as Ben Reade and guest starred on Relic Hunter in 2002. From 2003 to 2004, he appeared as Luc Johnston in the television series Tru Calling. In 2003, he appeared in the stage production Roulette, and in 2004, he appeared in an episode of North Shore. He made his film debut in Flightplan in 2005 and appeared in the film The Texas Chainsaw Massacre: The Beginning and the television film Amy Coyne in 2006. In 2007, he starred in the television series Traveler and appeared in a recurring role on Chuck from 2007 to 2009. He also appeared in the stage production Villa America in 2007.

In 2016, Bomer appeared in the films The Nice Guys and The Magnificent Seven, and he starred as Monroe Stahr in the television series The Last Tycoon. In 2017, he appeared in the films Walking Out and Anything. In 2018, he made his Broadway debut in the revival of The Boys in the Band, directed an episode of The Assassination of Gianni Versace: American Crime Story, and appeared in the films Jonathan, Papi Chulo, and Viper Club. He appeared in guest roles on Will & Grace from 2018 to 2019 and began starring as Negative Man in the television series Doom Patrol in 2019.
In 2020, he appeared in the television series The Sinner. In 2023, he executive produced and starred as Hawkins Fuller in the miniseries Fellow Travelers. He appeared in the film Maestro in 2023. In 2024, he was cast in the television series Mid-Century Modern, which premiered and was canceled in 2025. He is scheduled to co-star in the film Outcome, which began filming in 2024 and is expected to be released after its announcement in 2025.
