After graduating from Columbia College, Bryant toured with the musical improv group Baby Wants Candy and performed with iO Chicago, The Second City, and the Annoyance Theatre. She was a writer and ensemble member for Second City productions Sky's the Limit, Weather Permitting and We're All In This Room Together.
Bryant made her television debut as a featured player on Saturday Night Live on September 15, 2012, and was promoted to repertory player during her second season. In 2013, she had a recurring role on IFC's Comedy Bang! Bang! and appeared in The Amazing Spider-Man 2. She has guest-starred on shows including Broad City, Documentary Now!, The Awesomes, and Girls.
From 2016 to 2017, Bryant appeared as Alice in Louis C.K.'s Horace and Pete and voiced the main character D.D. Danger on the animated series Danger & Eggs. In 2019, she starred as the lead in Hulu's Shrill, which ran for three seasons through May 2021. She continued her role on Saturday Night Live through its 47th season (2021–2022).
Bryant voiced Emmy on the adult animated series Human Resources, which premiered in March 2022. She appeared on Broadway in All In: Comedy About Love in 2024. She is set to star in the upcoming Peacock dramedy Lonely Hearts Club.
