Lee graduated from Harvard University in 1994 with a degree in English literature and environmental studies. He made his Broadway debut in Urinetown in 2001 and went on to appear in numerous stage productions, including a musical adaptation of Hamlet in which he played Rosencrantz. In 2008, he received a Theatre World Award for Distinguished Performance for Yellow Face.

Lee began his television career in 2003 with an appearance as Dr. Mao on Sex and the City. His subsequent television credits include Law & Order (2006), Fringe (2008), The Unusuals (2009), Royal Pains (2009), White Collar (2009), Blue Bloods (2010), and NYC 22 (2012). From 2012 to 2017, he provided the voice of Hamato Yoshi/Splinter and additional characters on Teenage Mutant Ninja Turtles, appearing in 77 episodes. He also voiced a Korean Agent on Archer (2013).

In 2013, Lee began playing Job on Banshee, appearing in 31 episodes through 2016, as well as reprising the role in three episodes of Banshee Origins. His other television credits include Mako Tanida on The Blacklist (2014), Kee Mun on Hawaii Five-0 (2015), Reggie Woo on Bosch (2015–2018), Kenneth Park on Outcast (2017), Lei Kung on Iron Fist (2017–2018), and Norman Wong on NCIS: New Orleans (2019). From 2019 to 2023, he played Wang Chao in Warrior. He also voiced Ancestor and Oroku Saki/Shredder on Rise of the Teenage Mutant Ninja Turtles (2019–2020), played Toad in the second season of See (2021), and appeared as Wilson Lin in the miniseries DMZ (2022).

Since 2025, Lee has voiced White Rabbit in Devil May Cry and played Barney Choi in Your Friends & Neighbors. In 2026, he appeared in the miniseries East of Eden as Lee.

Lee's film credits include Saving Face (2004), as Raymond Wong; We Own the Night (2007), as an Emergency Services Driver; The Oranges (2011), as Henry; Exposed (2012), as Taki; Premium Rush (2012), as a Floor Manager; Identity Crisis (2016), as an Assassin; Mulan (2020), as the Village Magistrate; and The Monkey King (2023), in which he voiced the Jade Emperor and a Monk.

Lee has also provided voices for video games, including Grand Theft Auto: San Andreas (2004), as a Gangster; Grand Theft Auto: The Lost and Damned (2009), as the Crowd of Liberty City; Homefront (2011), as Colonel Jeong and Commander Park; Teenage Mutant Ninja Turtles (2013), as Hamato Yoshi/Splinter; Teenage Mutant Ninja Turtles: Danger of the Ooze (2014); and Nickelodeon All-Star Brawl 2 (2023).
