Plimpton began her acting career in the 1960s, creating the role of Crissy in the original 1967 Off-Broadway production of Hair. She continued in the role when Hair transferred to Broadway in 1968 and performed the song "Frank Mills" in both productions.
Plimpton took a leave from Hair to appear in the 1969 film Alice's Restaurant. She also appeared in Robert Downey Sr.'s Putney Swope in 1969, followed by Glen and Randa in 1971 and Hot Times in 1974. Her final film role was in Foreplay in 1975.
In 1986, Plimpton made a guest appearance on the television sitcom Throb. She subsequently retired from acting.

Casting has been announced for the Los Angeles LGBT Center's Lily Tomlin/Jane Wagner Cultural Arts Center's production of the iconic musical Hair. Under the direction of Kate Sullivan with musical direction by Jared Stein, there will be two preview performances on Wednesday, March 25, and Thursday, March 26, at 8pm. Opening is set for Friday, March 27, at 8pm in the Davidson/Valentini Theatre. The regular performance schedule will be Friday and Saturday at 8pm and Sunday at 7pm through April 26.
