I’m seeing TBV tomorrow (super excited!!), but after reading about people getting their bags taken up the Moulin Rogue thread, I was wondering, can I take a drawstring backpack with clothes in it into the theater with me?
I can't remember what happened when I saw TBV, but backpacks/small bags are generally fine in Shubert theaters. Security will quickly look in it as you enter the theatre. They're inconsistent about bottled beverages, so it's usually better not to bring those (sometimes my water bottle has made it in, other times I have to leave it on the sidewalk).
You shouldn't have any issue. I've brought in a backpack.

I saw the matinee show “The Band’s Visit” at the end of May on a Wednesday, had a standard-sized backpack, and they didn’t bother me or make me check it in. I had enough room under the stage in front of me to stow it away. I don’t remember how the seats were, but I’d probably make sure that it’s small enough to fit under them.
