Fun fact: found an old notebook of mine from about four years ago with a list for "Rocky Horror" scribbled in the margins; it was mostly crap but I did list Stark Sands (see above) AND I had Annaleigh Ashford as Columbia! The role she played (and slayed) in the (otherwise awful) TV remake!!!

Nicholas Barasch as Jack? I also think Nicholas Barasch would be a great Barnaby Tucker. I'd especially love to see him alongside Gavin Creel as Cornelius.
Does/can Cate Blanchett sing? I hope so! She'd be an amazing Desiree. She then could also reprise her role in the imaginary Carol: The Musical that lives in my head.

She sings well enough that she could sing Desiree. It's actually a very easy singing role, but a very challenging acting role.
In the upcoming revival of My Fair Lady at Lincoln Center, I would love to see a Frasier reunion with Kelsey Grammar and David Hyde Pierce alternating between Pickering and Higgins. Throw in John Mahoney as Dolittle if he can sing while you're at it. For Eliza, definitely Laura Benanti if she gets back in the game after having the baby or maybe Christy Altomore. If it has to be a Hollywood person, I think either Anne Hathaway or Anna Kendrick could pull the role off.
