- SEYMOUR KRELBORN — Male, 25–35. An insecure, put-upon florist's clerk and eventual hero. Genuine, well-meaning, taken for granted because of clumsy ways and poor social skills. Role will be understudied.
- AUDREY — Female, 25–35. Bleached-blond, Billie-Dawn-like, secret love of Seymour's life. Poor self-worth and education, incredibly good looks, sweet and vulnerable demeanor. Role will be understudied.
- MR. MUSHNIK — Male, 55–65. Seasoned owner of the failing East Side flower shop and Seymour's nosy boss. Profit-driven, greedy, manipulative. Role will be understudied.
- ORIN SCRIVELLO — Male, 30–40. An egotistical dentist with a passion for leather and sadistic tendencies. Audrey's abusive boyfriend, targeted by Seymour. Role will be understudied.
- VOICE OF THE PLANT — Any gender identity, 25–55. An offstage vocalist; voice of a conniving, street-smart villain. A funky R&B voice is best. Role will be understudied.
- CRYSTAL / RONNETTE / CHIFFON — Any gender identity, Black, 20–35. African-American street urchins acting as occasional Greek Chorus. Young, hip, smart. This trio has a grip on reality. Role will be understudied.
